How to extract region of interest (ROI), centroid (X, Y co-ordinates), pixel intensity, peak amplitudes from a stack of tiff/.tiff files of neural calcium imaging data?

I have an example neural calcium imaging tiff file (stack a tiff files from one video). I am interested to extract region of interest (ROI), centroid (X, Y co-ordinates), pixel intensity, peak amplitudes from a stack of tiff/.tiff files for my calcium imaging data from individual synaptic site.

Biological images are notoriously hard to analyze, but we may be able to help depending on what you want to do. So first read this link, then post your image. If it's quick and easy, someone may write code. If not you may get direction for how you can write it, or perhaps links to some papers on it or universities or consultants who may be able to write the app for you (if you don't want to write it yourself).

I still don't really know what you want measure. Do you want to measure the area of the green stuff? Or the area of the bright flash, whenever it occurs? What would you do with that image, if you had the MATLAB skills to do it?
