Detection of Frame Movement

Hi All, I have a very general question. I hope any person can help me. I have a set of medical videos and I need to find out difference between frames and based on that difference need to extract a bunch of results. Now as far as I have seen we need optical flow algorithms for this kind of extraction. I want to do this in matlab not in open cv or C# etc.

Please can anyone have an existing opticla flow algorithm which I can use to modify to my needs. Anyone can suggest reading material? or just point me in the direction where I should start?
