You must have the dSPACE TargetLink blockset installed. I've run into the same problem that I assume you are facing, models created by others contain "tllib" blocks and I don't have the license. Attempting to run a simulation in Simulink will always fail because it cannot find the reference "tllib" library.
If you have dSPACE licensed and installed then it is most likely an issue with your working path.
