MATLAB and optimisation tool automatically close while solving a large problem, I leave the program to run at night and in the day there are no programs running , I have tried the same optimisation for lesser number of variables and didn't face any problem in it.

Is it possible that your system goes into hybernation? Especially due to no user input? Historically there were a few cases where hibernation could kill MATLAB.

No , I have solved the exact same problem for a lower number of variables and in that case also MATLAB was running for about 14 hrs and it didn't got closed.
