Since asking this question, I have found that the National Instruments PCI 6351 does seem to work just fine using the PCI 6251 blocks. I am using the analog in and out, digital in and out, and one counter-timer for a quadrature encoder. This is good to know as it is hard to find motherboards with PCI slots anymore.
Is the National Instruments PCI-6351 board supported by Simulink Real-Time?

I am using Simulink Real-Time with an XPC target using a NI PCI-6251 for analog, digital, and counter-timer I/O. Can I build the next machine using a PCI-6351 board? It is not listed as supported hardware. I am hoping the PCI-6251 blocks work.

Hello Peter,
As you mentioned, the National Instruments PCI-6351 board is not listed in the supported hardware list below:
Therefore the hardware is not officially supported. It is possible that it may still work, but it is out of the expected workflow in Simulink Real-Time, and may cause issues.
